We tried RNA isolation from murine pancreas many times but we are getting degraded RNA. Kindly suggest precaution for the same.
What is your method of isolation?
Is the tissue fresh or biobanked?
How was the tissue homogenized?
Did you use a bioanalyzer?
Hi, you can communicate the detailed information about how to extract RNA from murine pancreas with the related technologists of commercial company or other researchers who do the same work as you! Maybe you can go to their lab to learn it! best wishes with you!
@Can Kiessling: We are using traditional method of RNA extraction by trizol reagent. Tissue were store in RNA later at -80C.We didn't use bioanalyzer and nor the tissue was homogenized.

Did you just put the tissue in Trizol without any homogenization?
Can you show us the image where you concluded the RNA was degraded?

@Can Kiessling: I have added representative gel image of RNA. We have acquire tissue (pancreas, liver and adipose tissue) from same mouse. We do not homogenize the tissue in the homogenizer, instead we manually chop/cut the tissue finely in the Trizol. No, the tissue was not fresh.
Lane 1: Pancreas, Lane 2: Liver & Liver 3: Adipose tissue.
