You need the rhizobium spore count between 108and 1010 CFU/mL and at least 15 to 20 mL , depends on your soil quality to mix your 100 g of soil .

Each

isolate of PGPR was grown overnight in Kings’ B

broth. Each broth was centrifuged at 12,000 rpm,

washed with phosphate buffer three times and then

pellets were dissolved in 0.1M phosphate buffer

(pH 7.0) and OD was adjusted to 1.2 before being

used for pot experiments. An OD of 1.2 was

equivalent to 2.0108 cfu ml1. The seeds for each

treatment were soaked for an hour in phosphate

saline buffer (PSB) containing the suspension of the

PGPR isolates to maintain a population of approximately

108 cfu seed1. In each pot, eight seeds (95%

germination) were sown at a depth of 5 cm. After

germination, five seedlings were maintained in

each pot.
