Hi, I spent lots of time to figure out ARA method to determine nitrogen fixation by free living in soil. However, I am not getting significance difference between ethylene peaks of control (no soil) and soil sample. It could be low biomass samples. Could suggest any specific protocol for low biomass soil.
I did ARA and 15N study with free living N2 fixing bacteria at IRRI, Philippines. Unfortunately, no significant N2 fixation was detected rather root surface area and carboxylase activity increased.
Shortly I read ARA doesnot detect N componds always surely. On the other hand there are also living endophytic bacterias in plants and in the root environment which promote plant growing as plant growth-promoting bacteria (PGPB) by nutrients and hormones and even often they control of plant pests which should be also considered. Dedection of such microbes could contribute to evidence of N flows and N sinks by plants.
