Hi, I tried 15N2 method to determine free living bacterial activity in soil samples at 2d, 4d and 8d incubation period. However, i have did not find considerable activity. Does incubation period influence activity. I am wondering to go with a short period of incubation like 2h, 4h, and 8h. Thanks....
what compound did you used for labelling? 15N2 means the gas material? If you used 15NO3-,15NH4+, you need to consider the transformation rate, I don't konw how to use N2 to label target in soil samples.
Placed 5g moisture soil in 20ml bottle, injected doubled labeled 1.5ml 15N2 gas at incubation 2d, 4d and 8d. Thanks.
That sound like you are relying on free living organisms that fix N2 gas.
Also a question: what exactly do you mean by 'double labelled'?
The first point is what kind of activity do you want to determine? global activity, N fixation activity, mineralisation activity, nitrification...?
