Being some years away from my last sets of ELISA experiments, now, does anyone have recommendations for a really reliable (i.e. good intra-sample reproducibililty and relevant dynamic sensitivity range) commercial kit currently available for detection of transforming growth factor beta in human CSF? I'd appreciate recommendations for both research grade and clinical grade options.
Although I have not used them for TGF-beta or CSF assays, I have used several R&D Systems ELISA kits and have always been pleased with their performance.
I have used the one from R&D Systems to quantify human TGF-beta release by Caco-2 cells. It went really well.
As mentioned below, the R&D kit is oaky, certainly for cell culture experiments and urine. However, assuming you are having to activate your samples to dissociate the TGFbeta from the lattency associated peptide, restoring the sample to the correct pH is critical. I'm also very wary of repeat freeze/thawing of TGFbeta.
eBiosciences h/mTGF-B Ready-set-go! kit sucks, since the diluent contains 10%FBS, that, by itself, has a large amount of TGF-Beta. Unless you are using your sups straight, I would not recommend using that kit. That also means you cannot use FBS in your cell cultures or your background may be too high.
