We commonly used 10% formaldehyde to fix tissue for pathological usage such as IHC or FISH.

It was reported that proteinase K or sonication+SDS can reduce the cross-linking level of the FFPE sample.

However, both proteinase treatment and sonication can damage the chromatin sate.

It was also reported a lysis buffer containing both ionic and nonionic detergents showed moderate effect of the damage.

Is there any other method that can reduce the cross-linking of the FFPE samples (for example, from 10% to 2%)?
