Can anyone suggest a good membrane permeable fluorescent dye for imaging intracellelar calcium which has emission either in red or far red region? Most of the good calcium sensitive dyes are green and the red dye from thermofisher is not compatible with my cells.
I am not an expert in Ca2+ visualization, but out of my experience with many fluorescent dyes I can tell, that commercial dyes are not always the best (aq. stability, photobleaching, etc.). I suggest to try to look around the state-of-art publications on calcium visualizations and speak directly with authors; they are usually happy to send a sample of their dyes and promote use of their own compounds. Or at least can help you.
Just out of my literature database came out few papers from the last years, e.g.: Angew. Chem. Int., 10.1002/anie.200701720 or 10.1002/anie.201210279 ; or xanthene-based: Article Synthesis of Practical Red Fluorescent Probe for Cytoplasmic...
(although it is not my own or my colleagues work, I have some experience with the silico-pyronins and would assure you, that they are possessing very good photophysical parameters as well as high solution- and photostability and could only recommned this class of dyes).Or maybe, please, write more details about your own issue and people can help better. Thank you for your suggestions. Most of the calcium probes I found in literature were either green or the ones from TF . I am studying calcium waves propagation in cells.
