We recently tried to bond -SH to magnetic-Au NPs (in citrate buffer) surface of Nanobead (250nm diameter).

The SH is at the end of a apatemer stucture (SH-apatmer-alexa488) The apatmer has 15 bases.

The SH- come with the format of -S-S- it was activated with TCEP solution (pH=7).

The activated -SH-apatmer-Beacon stucutre then mixed together with the magnetic-Au NPs and sonicate 30min + room temperature incubation (2 h). However, after the preparation, we concentrate the beads using a strong magnatic feid, we can see the aggregated beads under 20X, but just no GFP flouresent signal for these bead.

Our guess is either -S-S to -SH activation or -SH bonding to Au surface was falied.

Could anyone with this experise provide us some sugguestion?

Would citrate buffer that comes with magnetic-Au NPs block -SH bonding to Au surface?
