I’m trying to follow your published protocol in article entitle “ Quantification of Aloe Vera polysaccharides by O-Acetyl and UV-Vis spectrophotometry: First Action 2018.14”.

All parts are clear to me, but I’m not sure if I understand the total polysacharides calculation formula correctly:

As I’m working with callus, I scaled down the protocol and my starting material was 1 gr that I dissolved the extracted polysaccharids in 100 microliter of water and use all of it for quantification. My question is that in formula instead of “V” the total millimeters of sample solution, shall I put 0.1 ml or the total volume that I used for quantification after  adding alkaline hydroxyl amine hydrochloride, HCL and ferric chloride which was 0.7 ml.

Total Aloe Vera polysaccharids (%, w/w)

=[(A - b) /m] x [ V /W} x100

A= the absorbance of the test solution at 540 nm

b = the y-intercept of the calibration curve

m= the slope of the calibration curve

V = the total millimeters of sample solution

W = the weight of sample in milligrams
