I have a emergence question about the FACs in immunotherapy..

Any comment will be helpful..

I am going to go immunotherapy and this is my first project so I am having hard time to figure out what data I need to get.

I have kept searching for research article and read alot as well however, I think it is going to be much easier to get ideas if you guys can point out my questions. So please direct me and give me some basic info which will be so helpful to get better ideas and get better understand while I am reading research articles and for my research understanding.

my question is,

In immunotherapy, I need to use FACs to check if there is immune response, like T cell, NK, variety cytokines and etc..

can you guys point out what I need to search from FACs?

example, T cell/NK/IFN-gamma/ and so on..

so that I get the idea that what I can or need to find data from FACs.

Also,

Can you guys point me out what I need to search from other methods for other immune response ? like ELISA, or IHC... and so on.

I think IHC is for to see the T cell on the tumor tissue but I am not 100percent sure that this is only thing I can find in IHC..
