I used the qTOWER 3 qpcr machine of JENA company to carry out my qPCR assay, after 45 cycles, the results showed the amplification curve and the gene did has been amplificated, however, no CT values were shown in the data. What is the problem? Oh i found the reason is the threshold value, but the software cannot show the threshold button consistantly! How to solve this software problem?
Hi Kai-Lin Mao,
you can only see the thresholds by selecting "Calculate Ct". Otherwise it is just a normalized blot without a threshold value.
Select "Calculate Ct" (in the tab "Monitoring") and the color under the amplification plot and "Auto threshold" to get the CT values.

Stefan Koczan Thanks! Now i get it and it's because I did't click the "SCAN" button at the extention step, thus the machine didn't display the CT value.
