I am about to prepare the buffers (Tris 6,8/8,8, Laemmli Buffer, etc.) for my upcoming Western Blots and wondered if it is absolutely necessary to use distilled water or whether purified water would be "pure" enough for that matter.
Any advice and background explanation very much appreciated. Thanks a lot in advance!

Actually i usually use distilled water and i have never experience any problem with my buffer preparation with that when used for Western blot.If you use De-ionized water or distilled water i don't think is a problem at all.

It depends on what you mean by "purified". If it is a lab water system that gives you water with 18 Mega-Ohm resistance it is probably fine. I have never had a problem, but then I had the water filter rep test the incoming water and recommend the appropriate filter cartridges. Also, our water system depends on the building's Reverse Osmosis (RO) water as a source, so your water source will make a difference.
The gel running buffer and transfer buffers must be the correct pH WITHOUT correction, or the ion concentrations will be off and the solutions won't work correctly.
You might check what other researchers in your building have used, just to be on the safe side.

I agree with Adesola answer, in my experience, I tried bot, if you use De-ionized water or distilled water it should be fine. I remember multiple times I used tap water and my results were as good as when I used distilled or de-ionized water, but I do not recommend that, as per Jill comment, water source could be different.
