I have a serious problem with primary cell culture contamination with Pseudomonasaeruginosa. I culture porcine ovarian theca and granulosa cells, as well as corpus luteum. Which I cannot explain is that only theca and granolosa are infected. I belive that this contamination comes from animals. Has anyone similar problems? How to eliminate Pseudomas, usually used antibiotics (penicillin, streptomycin, gentamycin) do not work?
Pseudomonas aeruginosa may also originate from humid sources at it is a bug which tends to spread in sanitation, water plugs and so on. Make sure that there is no contamination source in your lab environment. Gentamicin and streptomycin may show activity against P. aeruginosa. However, many isolates, in particular veterinary isolates, have a number of antibiotic resistance genes. I would therefore recommend asking for a susceptibility test at a diagnostic lab. Based on the results of this susceptibility test you may choose an appropriate antibiotic for using in your cell culture.
Oliver Nolte, thank you for your answer. We did as you suggested, ans it seems that our bacteria is resistant to gentomycin and streptomycin. However, it is sensitive to tetracyclin, even in low doses.
We believe that the contamination is slaughtery origin.
