I have spheroids of mesenchymal cells (size between 300um - 400 um) and I wish to fixe them and dissociate them into single cells for posterior FACS. I tried to mechanically dissociate them with a P200 with additional vortexing of the cells but that proved to be insufficient. I was wondering if I need to add an enzymatic dissociation step. And if so, which would be the best enzyme to use?
