Protocol for primary cell culture establishment from normal rat pancreatic islet and how can I improve the number of generations of primary cell lines?

I want to establish primary cell culture from normal rat pancreas and want to get cell line of improved number of generations.

Tuğba Kiriş ....But i have facing problem in maintainig primary cell culture establish for long time...because the cell werte died earlier after 1 hour of isolation from rat pancreas.........please help me...what i can do establish cell culture for a long time in order to get the number of generation that can be further subjected for subculture and other desired experimentation purpose.....
