Protein isolation and purification

hi dear all

i am new in research feild and doing my PhD in biotechnolgy.i have to isolate some toxin form killer yeast.infact they are protein in nature i have to check their activity i.e its activitor and inhibitor of metal ions for this i need to make solution of metal salts please any body let me know how could i prepare the solution i mean what should be its formula to make soultion to check enzyme activity from stock solution and please also let me know which coulmn i have to use for my toxin i mean how should i select column for clorimateric assay. as you know we have so many cloumns for different dofferent proteins. thanks
