I am doing a protein assay for a membrane protein in 1% detergent LDAO (lauryldimethylamine oxide). I have tetraethylammonium ion (TEA+) in my buffer and it seems to interact with Lowry assay and form precipitate. I cannot use Bradford since it is not compatible. Can anyone help?
I have 8 Tyr and 2 Trp residues in my protein. I guess I can follow the absorptivity coefficient for Tyr and Trp at 280 (I think Trp is ~ 5500 and Tyr is ~1490) I will try measuring at A280 and also the LDAO blank, hopefully that nothing will come from the detergent. I should have noted that I also have 1% glycerol in my buffer, and wonder if that would interfere. Is there any particular protein assay in addition to this?
