Hi,
I am facing some problems in growing HelaS3 and K562 cells in suspension. Firstly, the media starts turning acidic (yellow) even at very low density of the cells i.e. ~0.3 million cells/ml. Right now I am growing cells in 175 cm2 flask using RPMI 1640 containing 10% FBS and Penstrep but I have to grow these cells in large spinner flasks. Secondly, the HelaS3 forms clumps which makes counting the cells difficult.
Try incubate your culture in the presence of carbon dioxide (~5%) , this can help to regulate the pH of your medium.

I think you must control the composition of your medium : contrôle the quality and quantity of bicarbonate, Also sera and antibiotic is very acid,you can balance this situation and increase the quantity of bicarbonate or changing for HEPES
To pass in the spinner you may be assure the good growth in flask
