I've been encountering a recurring problem with the YES (Yeast Estrogen Screen) test, specifically related to the development of an intense red color after adding CPRG.
I'm using disposable polypropylene plates, and ALL wells, including the solvent control (solvent, yeast, medium), and medium control (only yeast and medium) exhibit the same red color within 20 minutes after adding CPRG.
I've taken precautions by using single-use tips for each addition, and I've even prepared fresh media and solutions, but the issue persists. It almost seems like estrogenic compounds are present, giving a false positive result.
Has anyone else experienced a similar challenge with the YES test? If so, were you able to identify the root cause and find a solution? I would greatly appreciate any insights or suggestions to troubleshoot and resolve this issue.
