I've recently run into serious problems with my Western blot protein transfers to NC membrane. When I do the Ponceau stain, I get something as shown in the picture below - white blotches covering a good portion of the blot. Sometimes, the white area appears "oily"/hydrophobic (the rinse water sloughs off in the white areas specifically - although this is not always the case).

We have tried:

-changing to fresh buffers (running and transfer)

-new APS 10%

-new TEMED

-new acrylamide

-new Ponceau

-thicker sponges/more paper in the sandwich

-ice in the transfer apparatus to prevent heating

-soaking sponges and membrane before assembling transfer

Any ideas what could be causing the problem?
