Hi,

I was trying to generate spheroids from A375 cells by Pluronic F-127 precoating method. I used 1% F-127 in PBS to cover 12-well plate wells for 1 hour at room temperature, then rinsed the wells with PBS once, followed by cell seeding. Unfortunately, this didn't work and the cells got attached to the surface. Then I tried 10% F-127 PBS solution with the same procedure but this also failed to prevent adhesion. Now I am planning to skip the PBS washing part and just aspirate the F-127 off from the wells instead. Does anyone have any other suggestions?
