I usually use Quinacrine-Hoechst staining (QH staining) and Trypsin-Giemsa staining to analysis of human karyotype.
QH staining shows Q-banding of chromosomes, and Tripsin-Giemsa staining shows G-banding.
Generally, some one attempt to identify chromosomes by FISH analysis with DNA probe of ribosomal DNA, which shows special locations on each chromosome.
The first recommendation is Trypsin-Giemsa staining, because this method is very simple and widely used in all over the world and you can find many reference about almost of all animal and plant chromosome.
The second recommendation is QH staining, because this method is very very simple and the chromosome band is observed very clear. But, fluorescence microscope is required to observe the chromosome banding.
