Has anyone tried or aware of purification of P.falciparum proteins from the infected RBC culture lysate (endogenous protein and not any tagged version or heterologous expression)
Earlier I tried to isolate Plasmodium falciparum trophozoite stage protein i.e. hemozoin from digestive vacuole of the parasite. But you can check for specific protein you are interested in. Briefly, I did as given below: Spin whole blood and discard the plasma. Remaining cell population contains RBC, WBC and platelets and some protein. wash RBC by EDTA mixed normal saline three times (if you are doing in 1.5 ml Eppendorf). Then lyse cells by osmotic shock by sterile double distilled water. Use 1% SDS for washing residual hemoglobin after lysis of infected red cells from patients. Then lyze the vacuole and estimate the hemozoin content by spectrophotometry. This is what I did. But I am not sure which protein you are interested in.
