Generally I gave PEG treatment for 1 hour in 40C. It works fine. But for my work schedule it is better to use O/N PEG treatment. Is there any toxic or other effect of PEG which decreases the phage titer? Because in some source I found long time incubation is not better for phage isolation.
Firstly the temp used is 4 degrees and not 40 right? PEG functions to absorb water and aggregates phage particles together. It should be fine as phage particles are rather stubborn to harsh conditions. For your incubation time, Generally an hour would suffice but I don't see any harm to leave it o/n if you need to. During precipitation, if you suspect a loss in titer you could actually see if you lost any during the centrifugation wash step.
I have eventually left my phages overnight in PEG precipitation (on ice) without any problem to complete purification next day.
