I currently have a protein expression construct (for e coli expression) that is MBP tagged. However, the MBP tag sticks to my protein after cleavage. Therefore, I would like to express it as GST tagged. I would like cleavage to be via Factor Xa because it leaves no extra residues on my protein of interest.
It seems that pGEX-5X was historically a popular expression plasmid for GST-Xa fusion proteins, but a while ago the pGEX plasmids have been discontinued by GE (I am not sure but maybe they just got out of the plasmid business all together). Obviously, I can still get it in various forms from AddGene, but it makes me curious. Why has it been discontinued, and what are people using now for GST-Xa expression, or are there better options (that produce a protein with no extra residues after cleavage via inducible expression in bacteria))?
