I used PBS as my diluent instead of ddH2O when making sodium citrate buffer. Can anyone tell me what issues might occur during IHC epitope retrieval?

I think that it changes all. Ionic strength, pH, salt composition.... Therefore, your epitope maybe take a different conformation and would not be recognized by an specific Ab.
