Unfortunately, I couldn't find an example on internet, and needed to ask here. I am going to synthetically synthesise my fusion gene along with attL sites, although it will be costly. In this way, I can skip the gateway BP part and directly carry out LR between the double stranded linear gene fragment and a destination vector. Is there anyone who has done something similar before? What should I be careful on, especially while designing the gene fragment for synthesis? Thanks

I have not done the work so have no advice to offer. But I did use the following in a Google search and found numerous sources...

Thanks for your advice. I will look up on google using your key words. Just to update on the journey, because of the difficulty on synthesis of some genes, we received those in a plasmid rather than a fragment. We used them for LR gateway cloning and it worked expectedly (probably because of presence of attL sites).
