It's my first time culturing NK cells, not only mine but also the first time it is being done in my lab so I am kind of on my own. Does anyone have a step-by-step protocol they could share? I did my research and have been putting one together but I want to make sure I'm on the right track.

Hello Jasmin, We found that the simple addition of IL-2, as suggested by Sudeep, generated a significant activation of the TCRγδ+ population and population skewing from our density separated human PMBC cultures. We found as little as 20IU/ml rIL-2 changed the culture phenotype but did not evaluate natural killer cell markers eg CD56 or CD244 but would anticipate an expansion in this population too.
