NSCs are known to form floating neurosheres in 3D culture. What are the best culture media to be used for neurosphere culture of human NSCs? Are EGF and/or FGF required in the culture media for neurosphere culture?
I have read conflicting information about EGF and/or FGF requirement for the neurosphere culture. Most literature showed that EGF and FGF are required for neurosphere culture with the exception of cancerous NSCs from brain tumors (ie, glioblastoma). However, a neurosphere culture protocol from MilliporeSigma website recommends use of N2 media only without EGF or FGF growth factors. Expert advice would be really appreciated.
