We are looking for antibodies that efficiently label neurons for FACS analysis (possibly in the nucleus). We tried an antibody against NeuN from Chemicon but it did not work for us.
No specific neuronal type. We need to normalize quantifications done with a specific marker. Neuronal progenitors and/or terminally differentiated neurons would be fine.
You can try both early and late markers for normalization . Nestin being early marker for progenitors and Beta 3 tubulin for late (Terminal neurons). both markers are cytoplasmic not nucleus. chemicon anti bodies works fine for FACS. specificity to the source of your interest as to be verified before use.
did the procedure of staining NeuN involve permeablization? since its a nuclear localization 0.1% triton X for 30mins should do the trick.
mainly - Specificity of the antibody, Permeablization of the cells and cell type and the marker as to be verified
