I prepared liposome using the hydration rehydration method and after encapsulating my nanoparticle in the liposomes, zetapotential analysis does not show any result while DLS showed I had liposome of about 113nm in avg size. Expert advise on the zetasizer indicated the liposome could be degrading. I performed zeta potential analysis on ordinary liposome without nanoparticle and have -10.5mV. What could be causing the null result?
This is a common problem during zeta measurements. I would suggest to dilute your samples before zeta measurements. You can simply check by adding distilled water or buffer solution to your sample and then measuring zeta potential. This is not a big problem. However taking appropriate buffer for dilution is better as compared to distilled water. Also see if your samples are colored because it causes high scattering of laser 653 nm. It seems your phospholipids carry negative charges but your nanoparticle can neutralize them. One possible reason is that your nanoparticle has positive charge and make your loaded liposomes zwiterionic so the overall charge become zero. Or your nanoparticles can create an acidic pH condition after dissociation so make your phospholipids less dissociated. You can use a buffer to control the pH constant and select the suitable basic pH which makes the phospholipids have higher dissociation . The pH selection can be based on the pKa1 and pK2 of phospholipid species which you can find online.

@Jan. I am using nanosilver and my liposome is mainly DPPC. I didnt store the liposome. I only freeze a solution of the dissolve phospholipids and taw whenever I want to sonicate to make liposomes.
@Lisha. Thanks a lot. thats logical. my nanosilver is not really stable as I get bimodal peak for the zeta potential, one negative and the other positive in the same sample. I think this could be affecting it but not sure how to curb this and get a single highly negative r positive peak for the zeta potential.
your nanosilver will partly dissolve over time, i.e. generate silver ions interacting with dissociated phospholipids in your liposomes.

not familiar with details, however, it seems to me that some form of coating could minimized silver dissolution.
