I am working on developing a POC (lateral flow immunoassay) which uses blood as sample. The problem I face is that when the Test is initiated, the Nitrocellulose pad is having the blood particles and eventually developed the unsought red background. I feel the reason can be Hemolysis, that is causing the serum to red colored Hemoglobin. Is it right??

Treating sample pad with buffer system can help in sorting this problem? If so, May kindly advise the basic buffer composition.
