Please don't hesitate to ask me for me more details if required. Most publications on expression of recombinant therapeutic proteins via mircroalgae are based on growing them in photobioreactor. Why not Fermenters?
As you know fermenters are for heterotrophic cultivation conditions. Because there is limited numner of microalgae species whcih can grow in non-illuminated conditions this may be one of the reasons.
Because photosynthesis is the main driving force for energy and reproduction in a microalgae cell. So even if you recombinantly modify its metabolism you may still be dependent on photosynthesis unless it can grow in dark culture chambers while producing the recombinant protein or other chemicals.
This is my idea about the topic, they may be any other reasons.
Oral Vaccine for WSSV in shrimp using r-VP28 protein of WSSV
Recombinant proteins of white spot syndrome virus (WSSV) have been used to induce the resistance against WSSV infection through oral feeding. Different viral recombinant proteins have been tried and the results revealed that recombinant VP28 (r-VP28) protein was found to be more efficient and protect the shrimp from WSSV effectively.
Three different types of r-VP28 formulated diets i.e. Type A – feed coated with inactivated bacteria overexpressed with r-VP28; Type B – feed coated with pure form of r-VP28 protein; Type C – feed coated with nanoparticles encapsulated –VP28. In all the experiments, shrimp were fed with these three types of feed separately for 10 days (thrice per day) (r-VP28 concentration 50 g per gram of feed in all the experiments) and challenged on 11th day by feeding WSSV-infected meat orally. Survival rate ranged from 80 to 90% in WSSV-challenged shrimp in all experiments. Tissue distribution of r-VP28 protein and its retention in different organs of shrimp in all the groups were studied. Immune response of shrimp to r-VP28 protein in all treated groups was studied by analysing immunological parameters and up regulation of immune gene expression by real time PCR.
Based on the results of the Indian researchers, it clearly indicates the possibility of enhancing the resistance in shrimp against WSSV infection by immunostimulation using oral vaccine (r-VP28 protein of WSSV).
The efficiency of r-VP28 was further increased using our herbal extract. Large-scale production of r-VP28 has been standardized and it is ready for commercialization.
