Hi,
Has anyone ever used the services of outside companies (in the US or UK) for the generation of custom monoclonal antibodies (using mice) against recombinant proteins (there are no antibodies on the market for my proteins)?
If so, can anyone tell me their experiences - if they were successful or not, price, amount of immunogen needed?
I am trying to source a company but there are so many providing this service that I need to narrow down the field.

if there are no commercial Abs for your proteins, why not to try to make a polyclonal Ab (in rabbit, e.g.), first?
It requires less time to obtain a working reagent (first antiserum usually within 40-50 days), is cheaper, and easier to be caracterized. Last but not least, you can test the immunogenicity of your recombinant protein (which may then be used to achieve a monoclonal Ab).
I've a positive experience with Eurogentec, a worldwide supplier (I'm a customer of the Belgium site), but I never did monoclonals with them, if that's your specific need.
In the past years, I checked different companies, and I can say that there are no big differences between the immunization protocols, but for me are important also the relationship and the broad communication.
About the costs, there are several factors that impact on them, so you should directly ask for a quotation, but -generically- a polyclonal costs within (or around) 1000 euros, while a monoclonal Ab (by a commercial company) is from 5 to 10000 euros (or more), depending on the steps you're asking for (fusion, elisa screening, subcloning, supernatant production, purification, .... and so on).

Thanks for the advice. Unfortunately I need to get monoclonals because my proteins are mutants of each other and there will probably be a lot of cross reactivity with the polyclonals.
The sheer volume of companies that provides this service makes it difficult to choose one. I am only interested in the monoclonals at this point but I will look into Eurogentec. Of course, price and specificity are my two big concerns

Try looking into Abpro Labs. They specialize in custom antibody development. They have high success rates, and generate high quality antibodies the first time around. You should take a look at their site: www.abpro-labs.com

I don't know if you have already found someone to do this work but could I just say that I used to run the EMBL's monoclonal antibodies core facility which was very well known in Europe and very reliable.
