Has anyone done a titration curve looking at the amount of MOG peptide used to immunize C57BL/6 for EAE disease. I assume there is development of anergic T cells once a threshold is reached but I wonder how the EAE disease looks like at suboptimal doses of MOG. If anyone has tested it (with or without PTX) i'll be happy to hear how the results look

Hi Abbas,
we have tested titrations of MOG peptide or PTX respsectively [with leaving the other parameter constant starting from 200µg MOG peptide and 200ng PTX per injection] and monitored clinical EAE signs. I would NOT recommend to use less than 100 µg MOG peptide per mouse: Incidence decreased dramatically. Moreover, the usually synchronized EAE onset (of those few animals that got sick) spread tremendously (some animals got sick later than d20 [compared to d10 upon "standard" immunization]).
PTX titrations were in our hands much effective to modulate EAE: good correlation of PTX dose with EAE severity and incidence, but with minor influence on the synchronized onset.
However, we have not analyzed the frequency of anergic MOG-sepcific T cells - sorry
