Hello everyone,
I have found a new component that seems to generate tDCs (FACS, ELISA, WB and RTqPCR). But to be sure I do indeed have tDCs I am going to co-culture my DCs with naive T cells and see if I get Tregs.
However, is it best to use DCs and naiveTL from the same donor or from two different donors ? (I work on a cancer model) Moreover, do I need to add some kind on stimulus to the co-culture to help the reaction ?

The DCs and the naive T cells should be from the same donor so that there will be compatibility of MHC molecules which is essential for the activation of T cells
We don't add a stimulus for the co-culture . Just keep them in feeding medium for 7 days.
