I am using TaqMan® Advanced miRNA Assay.

For calibaration in qRT-PCR I use synthetic miRNA. The curve shape of synthetic miRNA looks different when we compare it with miRNA isolated from blood samples. Did you have the same experience or some explanation for this phenomenon?

All samples, including synthetic miRNAs, had the same total RNA input concentration for reverse transcription (1000 fmol/ul ~ 6,6 ng/ul).

Could anyone please help with the evaluation of the data?
