IT IS READY TO GO PRODUCT, WHERE THE PARTICULAR MIRNA WILL PRODUCE THE EFFECT ACCORDINGLY AND YOU CAN OBSERVE THE CHANGES WHAT YOU WANT TO. HERE, miScript MIRNA WILL DEFINITELY INCREASE OR DECREASE THE LEVEL OF PARTICULAR MIRNA WITHIN THE CELLS. THERE IN THE CELLS ENDOGENOUS LEVEL WILL BE ALSO THERE.

HERE IT WILL INCREASE OR DECREASE THE ENDOGENOUS LEVEL OF MIRNA, AND WILL BRING UP THE EFFECT.

POINT TO BE NOTED::: miScript mimic/inhibitor will give the effect of mirna without concerning about endogenous level means any particular mirna which u you are targeting its endogenous level is level is low or high, it doesn't effect, the product will give the overexpression and downregulation gross effect which might also include endogenous mirna contribution.

whereas, miRCURRY mimic/inhibitor will give answer of those mirna which is either very low or high in the cell. so if you know that what is the level of mirna within the cells then you prefer miRCURRY.

I will prefer firstly miScript, but it will depend on my applications & priorities.

As I said many times at researchgate, for transfection, you can also look for Polyplus Interferin transfection reagent which transfect mimics/inhibitor to cells very efficiently. I tried with Lipofectamine 3000 as many paper suggested but NOT WORKED AT ALL, it failed to transfect mimics/inhibitor to breast cancer cells, then many recommended Lipofectamine RNAi MAX. but I personally lost trust on lipfectamine product range for mirna work.

Last, YES you have set collateral negative control for the experiment.

I didn't get what do you mean by collateral negative control? I still don't know should i use All star negative control (that is usually used as a -ve control in sirna experiments) or i should buy the company-recommended -ve controls for both mimics and inhibitors ?

It will be better, if you go for recommended -ve controls for both mimics and inhibitors. You will be have accurate answer.
