i tried performing a CLP procedure to induce sepsis in mice and i followed all the instructions in advertised protocols and videos.
after performing the procedure, even a successfull and clean one, with minimal injury the mice did not wake up and even died right after.
any one has an idea what could go wrong?
According to your description, I think the most likely cause of death is excessive anesthesia. Can you describe the anesthesia plan in detail?
i used katamine and xylasine solution as writen in the literature and one fellow lab member used and worked (not with CLP but with other solution)
Hlo mam, there is much variability in anesthesia depth and mortality by ketamine95mg/kg and xylazine 7mg/kg
In mice ...or variable dosing ...may be for taht mice xylazine dose was higher that increases the depth of anesthesia with ketamine too , you may consider etomidate instead of ketmaine for inducing sepsis
