I'm currently performing my dissertation using Hs578T cells and I would like to try to perform the mammosphere assay. Has anyone ever worked with this cell line and did this assay? If so, can you tell me which is the best method to use and the main challegens I'll be facing giving that this is a particularly difficult cell line

I am not sure that i can help you but i can give you some examples what i tried.
Not all breast cancer cell lines are in the same amount capeable to form mammospheres. Some need to be detached from surfaces over a longer period of time (24-48h) to form floating mamosphers ore cell islets. Others need an increase in the viscosity oder fibrin in their media in different layers to form mammospheres. There are plenty of good papers out their but its more a try and error thing. Some companies provide very good supplements for your cell media to transform in a more natural habitat for your cells.
First you should determine what media is good for your cells and boosts proliferation the best. Then its on you to try to build a habitat or buy one.
