Hi,

For biodistribution, you should do a time course post injection. Ideally you could clone GFP on the AAV8 and follow the distribution with imaging . Once you have seen some distribution , harverst the organ and extrac RNA for RT-Q-PCR. You could also dissociate some of the tissue and do FACS analysis. We have published several manuscripts that you can find the protocols. However, our experiments were performed with Adeno. The principal is the same.

Thank you for you response. The AAV expresses mCherry. When performing the RT-PCR, are you suggesting to detect mCherry or should I detect other aspects (eg. promoter)? Thank you again for your insight.

I will try RT-Q-PCR for any gene that is easy to amplify in the AAV . Perhaps the REP . You can try the promoter ... The people that sell the AAV should have a good idea where area is easy to PCR.
