Hello all, these days I'm working on a hard study (to me). I use 3C protease, flagellin, and poly dadt to induce NLRP1, NLRC4, and AIM2 inflammasome activation, respectively, to see the compounds' selectivity. We have a positive drug MCC950, which is reported as an NLRP3-specific inhibitor, however, it (0.1-10uM) can inhibit NLRP1, NLRC4, and AIM2 activation. Is anyone working on this field? is it really solid that MCC950 is such a specific compound? And what could be the possible problem for my study, if I induced NLRP3 rather than NLRP1, NLRC4, or AIM2 activation?
