Making Pure Culture of Spread Plate from Mixed culture of Spread Plate

hi everyone. i have a question. If I have a streak plate of mixed culture and want to make a pure culture by spread plate technique for enumeration, how am I suppose to do so? Do I need to pick single colony from the plate and make serial dilution before spreading it on the plate? Is it possible to make serial dilution using a single colony?
