Hello, I need help in constructing my experiments in detecting the presence of a bacteria in my plants, via dna extraction

I have 10 treatments, with 6 replicates and 5 plants unit per replicate, totalling to 300 plants per experiment.

As for the dna extraction, I will be doing 1 destructive sampling from each treatment, monthly from the 6 replicates. So total of my biological replicates is 6 right?

I'm planning on selecting 3 different leaves part of the plants and combining them in one grinding, and performing the extraction on 2g of grinded powder separately (2 technical replicates) so 10 treatments x 6 replicates x 2 technical replicates = 120 extraction.

IS THIS CORRECT? Sorry for the lengthy post hope someone could help me! Thanks in advance!
