Hi All,
I am going to run BDNF ELISA in mouse brain tissue (prefrontal cortex and hippocampus). Can any one suggest a good lysis buffer and in what ratio should the protease inhibitor be mixed with lysis buffer during tissue homogenization? I am using Mouse BDNF ELISA kit from BIOMATIK. Thank you!

RIPA-buffer is commonly used (10-50 mM Tris, 150 mM NaCl, 0.1-0.5 mM desoxycholate, 1% Triton X100, 0.1-0.5% SDS) with phosphatase inhibitors (1 mM Na3VO4, 1 mM Na4P2O7), antibacterials (1 mM NaF) and protease inhibitors (start with commercial mixtures available as pill) as needed.
