I am doing extraction miRNA from sputum samples, by using extraction kit, but the problem is low concentration of miRNA. I have tried adding PBS 2-5 times into the sample before the extraction, but I still got low concentration. I can not get Sputasol or sputolysin in my country. Can anyone please suggest what should I do to get higher miRNA concentration?

Can I ask how are you measuring miRNA concentration? and what is the downstream analysis that you want to perform? We have used low concentration RNA for NGS applications with no problem at all.

Sergio Barberan-Soler , I am measuring miRNA concentration by using the nanophotometer, and I want to do the qPCR to compare the expression of the miRNAs in the sputum.
