I am looking for a sequence (s) l can use for extraction of MTB DNA from a sputum sample using either the Magvigen-Streptavdin-biotin DNA capture kit, or the MagVigen-Plasma DNA Select Kit. To my understanding the specific sequences are hybridized to magnetic beads which then pull out the DNA from solution. Any help will be much appreciated. Thank you in advance.
You can you GyrB gene sequence for specific identification of MTB. It will differentiate MTB to MOTT. GyrB gene sequence is long upto 2160 bp. First 700 bp having a specificity for MTB, rest of the sequence is homologous.
Why do you want to extract the Mycobacterium DNA? If you want to discriminate between positive and negative samples you can use lot of specific PCR or qPCR (including the atpE sequence for assessment of Mycobacterium spp in general). I'm not sure the divergence in the gyrB sequence between NTM and MTB will permit specific separation using DNA-DNA hybridation.
