Hello to all researchers, i need to do lentivirus titration and for this I need 10 fold serial dilution i.e 10^-1 to 10^-4...and i have my lentivirus stock stored with me...second thing is if i need to dilute my stock so which diluent i will be using distilled water,my routine growth media or serum free culture media?

You may thaw your lentiviral stock and prepare 10-fold serial dilution by diluting the lentiviral stock with routine growth media (complete media). Please do not vortex. You may add polybrene (if desired) to a final concentration of 6-10μg/ml. Polybrene is a cationic polymer that can greatly enhance the efficiency of lentiviral infection to mammalian cells by neutralizing the charge repulsion between virions and the cell surface, thereby increasing infection efficiency.
