Hi everyone,

I am trying to transduce my HEK293 cells with lentivirus containing a GFP construct.

Initially, I was getting a low GFP readout when assessing through FACS, so therefore we concentrated the virus using ultracentrifugation.

I used relatively arbitrary volumes of lentivirus to infect my HEK cells ( 6ul, 3ul, 0.3ul) and I saw the greatest transduction using the 0.3ul ( 11% ). Although this is still low, it was much higher than the other volumes used.

So would it be I would be required to titrate out using this volume of lentivirus ( 1:10 ) for example and try to see if serially diluting this out might improve the transduction efficiency using this relatively small stock volume?

Can the virus concentration be too high that it is not transducing the cells?

I did not see any cell toxicity in my HEKs, so I was a little stumped as to what was going on.

Any help would be appreciated as I relatively new to lentiviral work.
