Hello,

I am trying to find a minimal length of homology arms required to knock in small tags in the mouse ES cells. My tag is 207 bp long and wondering I need to have long homology arms. Plus, I would like to knock in this tag at several loci in the ES cells. Therefore amplifying the homology arms and then sequencing is quite time consuming. The strategy I would like to undertake is to synthesise dsDNA gene fragments consisting of the homology arms and the tag. Again, it's not possible to chemically synthesise long fragments. I am trying to find out the balance between the arms length and knock-in efficiency in ES cells. So that this would be a handy tool later on.
